富自体浓缩生长因子纤维蛋白提取液对MC3T3-E1成骨细胞增殖与分化的影响

Effects of Autologous Concentrated Growth Factors Extract on MC3T3-E1 Cells

摘要: 目的:提取富自体浓缩生长因子纤维蛋白(concentrated growth factors,CGF),研究CGF提取液对MC3T3-E1成骨细胞体外增殖、分化能力的影响。方法:制取CGF提取液,实验组采用CGF条件培养基,对照组为α-MEM完全培养基分别培养MC3T3-E1细胞,CCK-8法测定细胞增殖情况;ALP检测碱性磷酸酶活性;RT-PCR测定Runt相关转录因子-2(Runx2)和成骨细胞特异性转录因子(Osx)基因表达情况。结果:CCK-8显示各浓度CGF提取液有不同程度促增殖作用,其中10%CGF强于其他组;以10%CGF为实验浓度,ALP活性检测显示实验组3、5、7 d活性增强;RT-PCR结果显示实验组Runx2和Osx的基因表达水平均明显增高。结论:CGF释放的生长因子可促进MC3T3-E1成骨细胞增殖、分化。

Abstract: Objective: To evaluate the effect of the concentrated growth factors (CGF) extract on the proliferation and differentiation of MC3T3-E1 osteoblasts. Methods: The experimental group was cultured with α-MEM containing CGF, while the control group was α-MEM. The proliferation of the cell was detected by CCK-8, the activity of alkaline phosphatase was detected by ALP, and the expressions of runt-related transcription factor-2 (Runx2) and osterix (Osx) genes were quantified by real-time PCR. Results: CCK-8 showed that the experimental groups with different concentrations of CGF had different proliferation and the 10% CGF group was higher than other groups. When 10% CGF was used as experimental concentration, the ALP activity of experimental group was higher than that of control group after 3, 5, and 7d. RT-PCR results showed that the expressions of Runx2 and Osx in experimental group significantly enhanced. Conclusion: The growth factors released by CGF can promote the proliferation and differentiation of MC3T3-E1 osteoblasts.
