雌孕激素对人牙周膜细胞成骨分化影响的研究

Estrogen and Progesterone Modulates Proliferation and Differentiation of Human Periodontal Ligament Cells

摘要: 目的:研究雌孕激素对牙周膜细胞hPDLCs的成骨化的影响,探讨雌孕激素是否有协同作用。方法:原代培养hPDLCs,取第4~6代细胞用于实验。分为空白对照组,孕酮组,雌二醇组,孕酮组和雌二醇共同处理组。用含有50 nmol/L孕酮、50 nmol/L雌二醇的培养液培养hPDLCs 24 h或48 h后,采用MTT法检测在孕激素、雌激素作用下hPDLCs的增殖情况。通过Real-time PCR方法检测雌孕激素对牙周膜细胞hPDLCs中PLAP-1、OPG、ODF、Col-1和MMP-9等成骨分化相关基因表达的影响。茜素红染色定量分析雌孕激素对矿化hPDLCs细胞成骨分化的影响。结果:孕酮、雌激素干预hPDLCs后,细胞的增殖显著高于对照组(P<0.05),孕酮和雌激素共同处理组显著高于两者分别处理组(P<0.05)。与普通培养组相比较,孕酮和雌激素对成骨促进相关基因表达有显著的提高作用(P<0.05),两者共同作用促进相关基因更为明显。雌孕激素处理对矿化hPDLCs细胞成骨分化的影响高于对照组(P<0.05),两者联合显著高于单独处理组(P<0.01)。结论:孕酮和雌激素能够协同促进人牙周膜细胞的增殖和成骨分化

Abstract: Objective: To detect the proliferation and the expression of related genes of osteogenic differentiation in human periodontal ligament cells (hPDLCs) by estrogen and progesterone. Methods: The hPDLCs were isolated, cultured and stimulated with estrogen and/or progesterone for 24h or 72h. MTT assay was performed to assess the effects of estrogen and/or progesterone on cell proliferation. To quantify the effects of estrogen and/or progesterone on osteogenic differentiation of hPDLCs, real-time PCR was performed in estrogen and/or progesterone-treated hPDLCs. Results: Estrogen and/or progesterone could stimulate the proliferation of the hPDLCs, and promote the expression of PLAP-1, OPG, MGP, ODF, COL-1, and MMP-9 in hPDLCs. Furthermore, estrogen and/or progesterone exerted the synergistic effect in the promotion of cell proliferation and differentiation of hPDLCs. The effect of estrogen or progesterone treatment on osteoblast differentiation of mineralized hPDLCs cells was increased in contrast to the control group (P<0.05). The combined treatment group was significantly higher than the single treatment group (P<0.01). Conclusion: Estrogen and progesterone can synergistically stimulate the proliferation and differentiation of the hPDLCs.
