摘要: 目的:探讨磷酸二酯酶4D(PDE4D)在口腔鳞状细胞癌(OSCC)发生发展过程中的作用。方法:收集53例OSCC患者标本和5种OSCC细胞株,55例正常健康标本及2种正常口腔上皮细胞作为对照,利用qRT-PCR检测其中PDE4D mRNA表达水平;分别利用CCK-8,细胞划痕实验,流式细胞术和Western blot等技术检测PDE4D抑制剂Rolipram对SSC25细胞增殖、迁移、凋亡的影响。结果:OSCC患者组织中PDE4D mRNA相对水平显著高于正常对照组;OSCC细胞株中PDE4D mRNA相对水平显著高于正常细胞组,差异有显著统计学意义(P<0.01);且分化程度越高,PDE4D mRNA相对水平越高(P<0.05);Rolipram显著抑制SSC25细胞的增殖能力和迁移能力,促进细胞凋亡(P<0.05)。结论:PDE4D在OSCC发生发展过程中起关键调控作用。

Abstract: Objective: To investigate the role of phosphodiesterase 4D in the pathogenesis of oral squamous cell carcinoma (OSCC). Methods: Fifty-three OSCC patients and 5 OSCC cell lines were collected, 55 healthy adults and 2 normal oral epithelial cells were used as the control. The mRNA levels of PDE4D in these samples, the proliferation of SSC25, the migration abilities, and the cell apoptosis were examined by quantitative RT-PCR, CCK-8, wounding healing assay, and flow cytometry as well as western blot, respectively. Results: The levels of PDE4D mRNA in OSCC tissue and cell lines were significant higher than that in normal control group (P<0.01); the levels of PDE4D mRNA in OSCC tissue were positively correlated with the degree of OSCC differentiation (P<0.05); the abilities of cell proliferation and migration were significantly inhibited by Rolipram; Rolipram induced a significant apoptotic changes on SCC25 cells (P<0.05). Conclusion: PDE4D plays an important role in the OSCC pathogenesis.
