口腔鳞状细胞癌患者外周血microRNA的表达及预后意义

Expression and Prognostic Significance of microRNAs in Peripheral Blood of Patients with Oral Squamous Cell Carcinoma

摘要: 目的:筛选口腔鳞状细胞癌(OSCC)患者外周血差异表达的microRNA(miRNA),探讨其与临床参数及其预后意义。方法:利用qRT-PCR技术检测OSCC外周血中miRNA的变化,扩大样本量进行验证,并分析miRNA与OSCC临床病理指标和预后的关系。结果:总共检测84种miRNA,其中OSCC患者外周血中上调大于2倍的miRNA有8个;下调小于0.5倍的有18个。其中miR-222-3p和miR-423-5p在外周血的表达与pTNM分期和淋巴结转移显著相关(P<0.05)。miR-222-3p高表达和miR-423-5p低表达的患者生存期均较短(P<0.05)。多因素分析结果提示miR-222-3p高表达可作为OSCC患者生存的独立预后因素。结论:miR-222-3p和miR-423-5p在OSCC患者外周血中存在差异表达,并有望成为OSCC早期诊断和预测预后的生物标志物。

Abstract: Objective: To screen differentially expressed microRNAs (miRNAs) in peripheral blood of patients with oral squamous cell carcinoma (OSCC) by qRT-PCR, and further to explore the relationship between differentially expressed miRNAs and clinical parameters and prognosis. Methods: The changes of miRNA in peripheral blood of patients with OSCC were observed by qRT-PCR, and the relationship between miRNA and clinicopathological parameters and prognosis was analyzed. Results: A total of 84 kinds of miRNAs were detected, of which 8 were up-regulated more than twice in peripheral blood of patients with OSCC and 18 were down-regulated less than 0.5 times. The expression of miR-222-3p and miR-423-5p in peripheral blood was significantly correlated with pTNM stage and lymph node metastasis (P<0.05). The patients with high expression of miR-222-3p and low expression of miR-423-5p had a worse prognosis (P<0.05). Multivariate analysis showed that the overexpression of miR-222-3p was an independent prognostic factor for the OSCC patients. Conclusions: miR-222-3p and miR-423-5p were abnormally expressed in the peripheral blood in OSCC patients, and they were expected to be biomarkers for early diagnosis and predicting prognosis of OSCC.
