2种骨内植入材料界面的多核巨细胞分布与骨桥蛋白表达

Distribution of Multinucleated Giant Cell and Osteopontin Expression in Different Implant Materials-bone Interface

摘要: 目的:比较医用纯钛和聚甲基丙烯酸甲酯(PMMA)2种材料-骨界面多核巨细胞分布情况和骨桥蛋白表达情况。方法:将医用纯钛和PMMA制成的种植体各30枚植入30只SD大鼠的两侧股骨远端髁突内,分别于3、7、14、21和28 d时处死动物。制备脱钙石蜡切片,HE染色观察种植体-骨间隙愈合界面多核巨细胞的分布;免疫组织化学法检测种植体-骨界面骨桥蛋白的表达情况。结果:HE结果显示在5个时间点里均可以观察到种植体-骨界面有多核巨细胞的形成,3 d时达到高峰且PMMA组多核巨细胞的数量明显多于钛组(P<0.05);多核巨细胞数目随时间推移均呈现下降趋势,在21 d和28 d时无统计学差异。骨桥蛋白的表达在7d时达到高峰期,且医用纯钛组明显强于PMMA组(P<0.05)。随时间延长,骨桥蛋白在钛组和PMMA组的表达均逐渐减弱。结论:不同骨内植入材料影响其界面的多核巨细胞分布与骨桥蛋白的表达。

Abstract: Objective: To compare the distribution of multinucleated giant cells (MCG) and the expression of osteopontin (OPN) in bone-implant interface between medical pure titanium (Ti) and polymethylmethacrylate (PMMA). Methods: Thirty Ti and PMMA implants were implanted into the distal femoral condyle of 30 SD rats. The animals were sacrificed after 3, 7, 14, 21 and 28 days. The paraffin sections were prepared. The distribution of MCG in the implant-bone interface was observed by HE staining. The expression of OPN in the implant-bone interface was detected by immunohistochemistry. Results: HE staining showed that MCG could be observed in the implant-bone interface at all time points, and the peak appeared at the 3rd day. The number of MCG in PMMA group was significantly more than that of Ti group (P<0.05). The number of MCG decreased with time in both groups, and there was no statistical difference after 21 days and 28 days. The expression of OPN was peaked at 7 days, and the expression of OPN in Ti group was significantly stronger than that of PMMA group (P<0.05). With time, the expression of OPN was gradually weakened both in titanium group and PMMA group. Conclusion: The distribution of MCG and the expression of OPN may be related to different implant materials-bone interface.
