壳聚糖海藻酸钠复合海绵治疗大鼠干槽症的研究

Application of Chitosan Alginate Composite Sponge for Treatment of Dry Socket in Rats

摘要: 目的: 研究壳聚糖海藻酸钠复合海绵在大鼠干槽症中的治疗作用,以及与碘仿纱条对比,为临床应用提供依据。方法: 制作壳聚糖海藻酸钠复合海绵,行体外抑菌实验。将45只SD雄性大鼠拔除上颌第一磨牙感染干槽症,随机分为3组:A组空白组、B组壳聚糖海藻酸钠复合海绵组、C组:碘仿纱条组。观察并记录治疗后创口愈合情况,取上颌骨固定脱钙,HE染色进行组织病理学观察,免疫组化检测各组牙龈IL-6和IL-10的表达差异。结果: 壳聚糖海藻酸钠复合海绵结构均匀,有抑菌作用。与A组对比,肉眼观察、组织病理学观察及免疫组化结果均显示B组愈合良好,炎症控制(P<0.05),且与C组无统计学差异(P>0.05)。结论: 壳聚糖海藻酸钠复合海绵能够抗菌、消炎,治疗大鼠干槽症,且能达到与碘仿纱条相当的治疗效果。

Abstract: Objective: To study the therapeutic effect of chitosan alginate sponge in dry socket of rats and to provide basis for clinical application. Methods: Chitosan sodium alginate composite sponge was prepared and performed in vitro bacteriostatic test. 45 SD male rats were extracted the maxillary first molar and infected with dry socket, which were randomly divided into three groups: Group A, blank group, Group B, chitosan alginate composite sponge group, and Group C, iodine imitation yarn group. The wound healing was observed and recorded after treatment. The maxillary bone was removed and decalcified. HE staining was used for histopathological observation, and the expression of gingival IL-6 and IL-10 was detected by immunohistochemistry. Results: Compared with Group A, Group B had better healing and inflammation control observed by naked eye, histopathological analysis, and immunohistochemistry analysis (P<0.05), and there was no difference between Group B and Group C (P>0.05). Conclusion: Chitosan sodium alginate composite sponge has antibacterial and anti-inflammatory property. The treatment of rat dry socket with sponge can achieve the same therapeutic effect as iodine imitation yarn.
