PTH-FC对大鼠骨髓间充质干细胞增殖及骨向分化的影响

Effects of PTH-FC on Proliferation and Osteogenic Differentiation of Rat Bone Marrow Mesenchymal Stem Cells

摘要: 目的:评价甲状旁腺激素融合蛋白PTH-FC(parathyroid hormone -Fc)对大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)骨向分化的作用,筛选出PTH-FC诱导BMSCs最佳的作用时间。方法:采用全骨髓贴壁法分离培养大鼠BMSCs并进行鉴定,以1.0×10-9 mol/L PTH-FC成骨诱导液诱导BMSCs作用6 h(间歇性给药组)和48 h(持续性给药组)通过噻唑蓝(methylthiazolyldiphenyl-tetrazolium bromide, MTT)、碱性磷酸酶(alkaline phosphatase,ALP)、茜素红染色观察PTH-FC对BMSCs增殖和骨向分化的作用。结果:含1.0×10-9 mol/L PTH-FC的成骨培养液作用6 h间歇性给药组与其他组比较,ALP和茜素红染色结果显著高于其他组;MTT结果显示各实验组间比较差异无统计学意义。结论:1.0×10-9 mol/L PTH-FC作用BMSCs 6h的间歇性给药组具有最佳的骨向诱导能力,这提示PTH-FC作为骨诱导生长因子联合BMSCs对骨组织缺损的修复是可行的。

Abstract: Objective: To evaluate the effects of PTH-FC on osteogenic differentiation of rat bone marrow mesenchymal stem cells (BMSCs), and to screen out the optimal time of BMSCs induced by PTH-FC. Methods: BMSCs from rats were isolated and cultured in vitro using the whole bone marrow adherence method and identified. BMSCs was induced with PTH-FC (10-9mol/L) osteogenic inducer for 6h (intermittent administration) and 48h (continuous administration). The best proliferation promoting and osteogenic differentiation promoting effect of PTH-FC on BMSCs was evaluated by means of MTT, ALP, and alizarin red staining. Results: ALP and alizarin red staining in the osteogenic culture medium of PTH-FC (10-9mol/L) which acted on BMSCs for 6h were significantly higher than those of other groups (P<0.05), however, the cell proliferation result was not significant different between the experimental groups. Conclusion: The combination of PTH-FC, which is used as a bone induced growth factor, with BMSCs to repair bone tissue defects is feasible.
