一种新型牙骨质细胞系的矿化特性研究

Mineralization of A Novel Immortalized Cementocyte Cell Line: A Pilot Study.

摘要: 目的: 探究一种新型的牙骨质样细胞系IDG-CM6的矿化特性。方法: 体内实验采用3月龄C57BL/6雄性小鼠下颌第一磨牙近中根的细胞牙骨质和牙槽骨扫描电镜(scanning electron microscope,SEM)及共聚焦显微拉曼光谱为参考;体外培养以骨细胞系IDG-SW3细胞为对照,对矿化3、7、14、21和28 d的IDG-CM6细胞及IDG-SW3细胞进行茜素红染色、SEM及共聚焦显微拉曼光谱扫描,研究其矿化特性。结果: 同一时间节点的茜素红染色结果显示,两种细胞均在培养中出现明显矿化,IDG-CM6细胞钙离子沉积更加明显。SEM及能谱仪(energy dispersive spectrometer,EDS)分析显示,两种细胞的Ca/P比差异无统计学意义,与体内实验结果一致。拉曼分析结果显示相同矿化节点,IDG-CM6细胞的PO43-/CH2(矿化成分/有机基质)比值显著低于IDG-SW3细胞,这与体内细胞牙骨质的PO43-/CH2比值(1.76)明显低于牙槽骨(4.76)一致。结论: 新型牙骨质细胞系IDG-CM6细胞可能是研究牙骨质矿化特征及生物学特性的理想体外模型。

Abstract: Objective: To investigate the mineralization of a novel murine immortalized cementocyte-like cell line IDG-CM6.Methods: In vivo, three-month-old C57BL/6 male mice were sacrificed to collect the cellular cementum and alveolar bone around the mesial root of the first mandibular molar for scanning electron microscopy (SEM) and Raman spectroscopy.In vitro, IDG-CM6 cells were cultured for 3, 7, 14, 21, and 28 days to compare with osteoblast/osteocyte cell line IDG-SW3.Alizarin red staining, SEM, and Raman spectroscopy were used to analyze the cells.Results: Both two cells mineralized during the cell culture.However, the alizarin red staining showed significant calcium deposition in IDG-CM6 cells compared with IDG-SW3 cells at the same time course.SEM showed that there was no obvious difference in Ca/P ratio between two kinds of cells, consistent with that of the cellular cementum and alveolar bone.Raman analysis indicated that the PO43-/CH2 of IDG-CM6 cells was significantly lower than that of IDG-SW3 cells, consistent with the markedly lower PO43-/CH2 (1.76) of cellular cementum than that of alveolar bone (4.76).Conclusion: IDG-CM6 cells might be an ideal model to study the mineralization and biological characteristics of cementum.
