口腔血链球菌提取物对SKOV3细胞的抑制作用

Inhibitory Effect of Oral Streptococcus Sanguis Extract on SKOV3 Cells

摘要: 目的: 血链球菌提取物XLS是从口腔血链球菌中提取的具有一定生物活性作用的细菌素样物质。本研究旨在探讨XLS对卵巢癌(ovarian cancer,OC)SKOV3细胞系的抑制作用及其可能的分子机制,为后续对XLS的相关研究和应用提供理论依据。方法: (1)采用CCK-8法检测XLS和紫杉醇分别对SKOV3细胞活性的影响;(2)通过克隆形成、Transwell、细胞划痕实验检测XLS对细胞增殖、侵袭和迁移的影响;(3)使用Western blot和实时荧光定量聚合酶链反应(quantitative real-time polymerase chain reaction,qPCR)分析XLS对磷脂酰肌醇3激酶/蛋白激酶B(phosphatidylinositol 3 kinase/protein kinase B,PI3K/Akt)通路及相关蛋白表达的调控作用;(4)检测线粒体膜电位、活性氧(reactive oxygen species,ROS)和Ca2+水平以探讨XLS诱导的线粒体凋亡。结果: (1)经XLS处理后SKOV3细胞的活力随XLS浓度的增加而下降;(2)XLS表现出浓度依赖性抑制SKOV3细胞增殖,侵袭和迁移;(3)本实验中XLS能抑制PI3K/Akt信号通路的激活,调节相关基因蛋白的表达,诱导细胞凋亡;(4)XLS可降低线粒体膜电位,增加氧化应激和Ca2+失衡,进一步破坏细胞结构和功能。结论: XLS能够抑制SKOV3细胞的增殖、侵袭和迁移,诱导凋亡,表现出显著抑制SKOV3细胞的潜力。

Abstract: Objective: To investigate the inhibitory effect of Xue Lian Su (XLS), a kind of bacteriocin-like substance extracted from oral streptococcus sanguis, on ovarian cancer (OC) SKOV3 cell line and its possible molecular mechanism. Methods: The effects of XLS and paclitaxel on the activity of SKOV3 cells were detected by CCK-8 method. The effects of XLS on cell proliferation, invasion, and migration were detected by colony formation, Transwell, and cell scratch. Western blot and quantitative real-time polymerase chain reaction (qPCR) were used to analyze the regulation of XLS on phosphatidylinositol 3 kinase/protein kinase B (PI3K/Akt) pathway and related protein expression. Mitochondrial membrane potential, reactive oxygen species (ROS), and Ca2+ levels were detected to investigate XLS-induced mitochondrial apoptosis. Results: The viability of SKOV3 cells treated with XLS decreased with the increase of XLS concentration. XLS inhibited the proliferation, invasion, and migration of SKOV3 cells in a concentration-dependent manner. XLS inhibited the activation of PI3K/Akt signaling pathway, regulated the expression of related gene proteins, and induced apoptosis. XLS could reduce mitochondrial membrane potential, increase oxidative stress and Ca2+ imbalance, and further destroy cell structure and function. Conclusion: XLS can inhibit the proliferation, invasion, and migration of SKOV3 cells and induce apoptosis, showing the potential to significantly inhibit SKOV3 cells.
