正畸力作用下压力侧Wnt10b、RANKL在牙周组织中的表达

Expression of Wnt10b and RANKL in the Pressure Site of Periodontal Tissue under Orthodontic Force.

摘要: 目的: 研究大鼠在正畸力作用下压力侧wnt10b、RANKL 在牙周组织中的表达变化。方法: 将42只健康雄性SD大鼠随机分为7组,即0 h、6 h、12 h、24 h、5 d、7 d、14 d组,使用镍钛拉簧对第一磨牙向近中施加50 g正畸力,以0 h为对照组,每组处死6只。应用实时定量PCR(RT-PCR)对wnt10b、RANKL的mRNA表达进行检测。结果: Real-time PCR结果显示,与对照组相比,实验组压力侧牙周组织中Wnt10b、RANKL mRNA均有表达,其中wnt10b于加力初期表达受抑制,随后升高,于5 d达高峰随后下降,14 d恢复至正常水平。RANKL表达随加力时间延长而逐渐升高,7 d达到高峰,随后下降。结论: Wnt10b、RANKL参与正畸加力作用下的牙周组织改建。

Abstract: Objective: To investigate the changes of RANKL and wnt10b in the periodontal tissue during orthodontic tooth movement. Methods: 〖WT5”BZ〗Forty two healthy male SD rats were randomly divided into 7 groups according to the application loaded time points of 0h, 6h, 12h, 24h, 5d, 7d, and 14d. The group of 0h served as control and the Nickel-titanium closed-coils springs were used to produce 50g power to drag the first molars mesially. Real-time PCR (RT-PCR) technology was used to detect the expression of Wnt10b and RANKL. Results: 〖WT5”BZ〗RANKL and Wnt10b were both expressed in the pressure site in comparison with the control group according to the results of the RT-PCR. The expression of wnt10b mRNA was suppressed at the early stage, then increased, and reached the peak after 5-7 days. It returned to the normal level after 14 days. The expression of RANKL elevated at first, then peaked after 5-7 days, and decreased. Conclusion: 〖WT5”BZ〗Wnt10b and RANKL participated in periodontal tissue remodeling under the action of the orthodontic force.
