AE-PDT对裸鼠舌鳞癌治疗效果的初步探讨

Preliminary Study on Therapeutic Effect of AE-PDT on Tongue Squamous Cell Carcinoma in Nude Mice.

摘要: 目的: 探讨芦荟大黄素(aloe-emodin,AE)介导的光动力疗法(photodynamic therapy,PDT)对人舌鳞癌治疗的疗效,为临床提供理论依据。方法: 将人舌鳞癌Tca-8113细胞植入裸鼠皮下,随机分4组(对照组、光照组、AE组和PDT组),每组9只。治疗后定期测量肿瘤体积,14 d后各组处死3只,剖瘤称重计算抑瘤率;并行病理形态学观察;其余6只观察生存时间,计算生命延长率。结果: 各处理组肿瘤体积均小于对照组,PDT组肿瘤体积变化明显(P<0.05);光照组、AE组、PDT组抑瘤率分别为:4.76%、20.24%、77.40%,PDT组抑瘤效果明显优于其他组; HE染色可见PDT组肿瘤细胞坏死及空泡变性,对照组及光照组无明显改变;各处理组均能延长小鼠生存时间,但以PDT组生存期最长、生命延长率最高。结论: AE-PDT对舌鳞癌肿瘤的生长具有抑制作用,并可延长肿瘤小鼠的生存时间。

Abstract: Objective: To investigate the efficacy of aloe-emodin (AE)-mediated photodynamic therapy (PDT) in the treatment of human tongue squamous cell carcinoma, and to provide a theoretical basis for clinical treatment. Methods: Tca-8113 cells were implanted into nude mice subcutaneously in 4 groups (control group, light group, AE group, and PDT group). After treatment, the tumor volume was measured periodically. After 14 days, three mice were sacrificed and the tumor growth rate was calculated in each group. The survival time was observed and the life extension rate was calculated in the remaining mice. Results: The tumor volume of PDT group was significantly higher than that of control group (P<0.05). The tumor inhibition rates of light group, AE group, and PDT group were 4.76%, 20.24%, and 77.40% respectively. The results showed the tumor necrosis and vacuolar degeneration in PDT group which were not obviously detected in the control group and the light group. All treatments could prolong the survival time of the mice, but the PDT group exhibited the longest survival time and the highest rate of extension. Conclusion: AE-PDT can inhibit the growth of tongue squamous cell carcinoma and prolong the survival time of tumor mice.
