摘要: 目的: 探讨人脐带间充质干细胞(human umbilical cord blood mesenchymal stem cells,HUC-MSCs)对舌鳞状细胞癌Cal-27细胞侵袭和迁移能力的影响。方法: 细胞划痕实验检测HUC-MSCs分泌的细胞因子对Cal-27细胞迁移能力的影响;Transwell小室比较共培养前后Cal-27细胞的侵袭和迁移能力;逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测共培养前后Cal-27细胞的侵袭、迁移相关基因的表达水平;Western blot技术检测肿瘤侵袭转移相关蛋白分子基质金属蛋白酶(matrix metalloproteinase,MMP)-2和MMP-9的蛋白表达的情况。结果: 条件培养基培养的Cal-27细胞的迁移距离短于对照组;共培养后Cal-27细胞的侵袭和迁移细胞数目减少;侵袭迁移相关基因、蛋白的表达水平下调。结论: HUC-MSCs与Cal-27细胞共培养可以抑制Cal-27细胞的侵袭和迁移。

Abstract: Objective: To investigate the effect of human umbilical cord mesenchymal stem cells (HUC-MSCs) on the invasion and migration of Cal-27 cells in tongue squamous cell carcinoma. Methods: HUC-MSCs conditioned medium was prepared. Cal-27 cells were cultured and the effect of HUC-MSCs secreted factors on Cal-27 cell migration was detected by cell scratch test. The invasiveness and migration of Cal-27 cells before and after co-culture were compared in Transwell cells. Expression levels of genes related to invasion and migration of Cal-27 cells were detected by RT-PCR before and after co-culture. The changes of MMP2 and MMP9 protein expression in tumor invasion and metastasis-related protein molecules were detected by western blot. Results: The migration distance of Cal-27 cells was shorter than that of the control group. After co-culture, the number of invasion and migration of Cal-27 cells decreased. The expression levels of aggression-related genes and proteins were down-regulated. Conclusion: The co-culture of HUC-MSCs and Cal-27 cells can inhibit the invasion and migration of Cal-27 cells.
