饰胶蛋白聚糖与口腔鳞状细胞癌发生发展的相关性研究

Correlation between Decorin and Oral Squamous Cell Carcinoma

摘要: 目的:探讨饰胶蛋白聚糖(Decorin, DCN)在口腔鳞状细胞癌(oral squamous cell carcinoma, OSCC)中的表达和意义,及DCN影响OSCC发生发展的可能机制。方法:采用免疫组织化学法检测DCN、表皮生长因子受体(epidermal growth factor receptor, EGFR)、致癌基因C-Myc和细胞周期蛋白依赖性激酶抑制剂(cyclin dependent kinase inhibitor, P21)四种因子在OSCC组织和正常口腔黏膜组织中的表达;用脂质体法将DCN质粒转入HSC-3细胞中,采用Real time PCR和Western blot检测DCN过表达对EGFR、C-Myc和P21表达的影响。结果:随着OSCC分化程度的降低,DCN蛋白和P21蛋白的阳性表达率逐渐降低,EGFR蛋白和C-Myc蛋白的阳性表达率逐渐升高(P<0.05);与空载体组和对照组相比,转染组EGFR和C-Myc mRNA表达量和蛋白表达水平皆降低,P21 mRNA表达量和蛋白表达水平皆升高(P<0.05)结论:DCN与OSCC的发生、发展过程紧密相关,其对OSCC的作用可能通过调节EGFR、C-Myc和P21实现。

Abstract: Objective: To investigate the expression and significance of Decorin (DCN) in oral squamous cell carcinoma (OSCC), and the possible mechanism by which DCN affects the development of OSCC. Methods: The expressions of the DCN, epidermal growth factor receptor (EGFR), oncogene C-Myc, and cyclin dependent kinase inhibitor (P21) in OSCC tissues and normal gingiva tissues were detected by immunohistochemistry. After being transfected with DCN plasmid, the expressions of EGFR, C-Myc, and P21 in HSC-3 cells were detected by Real time PCR and Western blot. Results: With the decreasing of differentiation degree of OSCC, the positive expressions of DCN and P21 proteins were decreased gradually, while the EGFR and C-Myc proteins were increased gradually (P<0.05). Compared with HK group and control group, the mRNA and protein levels of EGFR and C-Myc in transfection group were decreased, while the mRNA and protein level of P21 was increased (P<0.05). Conclusion: DCN is related to the occurrence and development of OSCC, which might reach the goal by regulating the expressions of EGFR, C-Myc, and P21.
