口腔鳞状细胞癌患者年龄分布与肿瘤分子标记物的关系研究

Relationship between Distribution of Age and Tumor Biomarkers in Patients with Oral Squamous Cell Carcinoma

摘要: 目的:研究口腔鳞状细胞癌患者的年龄分布与肿瘤分子标记物的关系,为口腔鳞状细胞癌的诊断和治疗提供依据。方法:收集TCGA数据库头颈鳞状细胞癌中舌、颊、牙槽嵴、口底、唇、舌根、腭等部位发生的口腔鳞状细胞癌患者的数据。收集2018年1月~2018年12月武汉大学口腔医院口腔鳞状细胞癌患者数据。按不同年龄段分析患者的病例数,以及口腔鳞状细胞癌重要分子标记物p53、CCND1、EGFR和SOX2的表达与患者年龄的关系。进一步分析了p53的基因突变与患者年龄和p53 mRNA表达水平的关系。结果:口腔鳞状细胞癌主要发生在51~70岁的年龄段;CCND1和SOX2的表达在各年龄段没有显著差别;EGFR的表达虽然在各年龄段没有统计学的显著差别(P=0.107),但小于51岁组的表达水平要高于其它组1.5~1.7倍;p53的表达在各年龄段有显著差别,其中61~70岁组的表达量显著低于其它年龄组;p53基因突变率在各个年龄段没有显著差别;携带p53突变的口腔鳞状细胞癌患者的p53 mRNA表达水平显著低于p53未突变的患者。结论:口腔鳞状细胞癌高发于51~70岁年龄段,p53的表达在61~70岁年龄段的患者中显著降低。

Abstract: Objective: To investigate the relationship between the distribution of age and tumor biomarkers in patients with oral squamous cell carcinoma. Methods: The data of patients with oral squamous cell carcinoma were collected from TCGA database and Hospital of Stomatology, Wuhan University. The correlation analysis was conducted between the distribution of age and the expression of p53, CCND1, EGFR, and SOX2. Further correlation analysis was conducted between p53 gene mutation and the distribution of age. Results: Patients with age between 51 and 70 years were the most affected population in oral squamous cell carcinoma. No significant difference on the expression of CCND1, EGFR, and SOX2 was found among different age groups. However, a 1.5- to 1.7-fold increased expression of EGFR was observed in patients younger than 51 years old. Patients with age between 61 to 70 years showed significantly lower expression of p53 than other age groups. No significant difference on the incidence of p53 mutation was found among age groups. However, patients with wild type p53 had significantly higher levels of p53 mRNA than those with mutant p53. Conclusion: Patients with age between 51 and 70 years are the most affected population in oral squamous cell carcinoma. Patients with age between 61 to 70 years showed significantly lower expression of p53 than other age groups.
