基于GEO数据挖掘与网络药理学探讨活血化瘀方治疗口腔扁平苔藓的机制研究

Study on Mechanism of Huoxue Huayu Prescription in the Treatment of Oral Lichen Planus Based on GEO Data Mining and Network Pharmacology

摘要: 基于GEO数据库挖掘OLP的潜在治疗靶点,基于网络药理学分析活血化瘀方治疗OLP的主要化学成分、潜在作用靶点与治疗机制。采用R语言爬取GEO数据库中GSE52130数据集,通过差异分析得出OLP的异常表达基因;基于TCMSP数据库获取活血化瘀方中相关化学成分与靶点,利用Perl、Cytoscape软件和String绘制该药方成分-靶点网络与蛋白-蛋白互作网络;通过R语言完成关键靶基因的基因本体论(GO)分析与基因组百科全书(KEGG)分析。结果表明:活血化瘀方治疗OLP的关键化学成分共28个,包括槲皮素、山奈酚、豆甾醇等;核心作用靶点共13个,包括IL-1β、PTGS2(COX-2)、MAO等。KEGG分析发现活血化瘀方可影响神经递质代谢、巨细胞病毒感染、AGE-RAGE等通路,为后续中医药治疗口腔扁平苔藓的深入研究提供理论参考。

Abstract: Objective: To study the key components, potential targets, and mechanisms of Huoxue Huayu prescription in the treatment of oral lichen planus based on GEO data mining and network pharmacology. Methods: The GSE52130 data set in GEO database was crawled by R language, and differential therapeutic targets of OLP were analyzed. The chemical constituents and targets of Huoxue Huayu prescription were obtained based on TCMSP database, and the composition-target network and protein-protein interaction network were plotted using Perl software, Cytoscape software, and String. The GO and KEGG analysis of key target genes were completed by R language. Results: There were 28 key chemical components of Huoxue Huyu prescription in OLP treatment, including quercetin, kaempferol, stigmasterol, etc. And 13 core targets were found, including IL-1β, PTGS2 (COX-2), MAO, et al. KEGG analysis showed that Huoxue Huayu prescription could affect neurotransmitter metabolism, cytomegalovirus infection, and AGE-RAGE pathways. Conclusion: The result provided theoretical reference for the subsequent in depth study of TCM treatment of OLP.
