贮存条件对喷砂酸蚀钛种植体表面成骨细胞活性的影响

Effect of Storage Condition on the Osteoblasts Activity on Sandblasted and Acid-etched Titanium Surfaces.

摘要: 目的:研究不同贮存条件对与喷砂酸蚀钛种植体表面成骨细胞黏附、增殖等影响。方法:采用喷砂酸蚀技术对钛片进行改性,并将钛片分别放入组A密闭容器、组B双蒸水(ddH2O)、组C氢氧化钠(NaOH)中保存4周。将成骨细胞接种到钛片表面,通过细胞形态观察,黏附增殖检测、碱性磷酸酶活性检测,分析3种贮存方法对种植体的影响。结果:在3 h、1 d、4 d、7 d,成骨细胞在钛片上黏附增殖具有统计学意义(p<0.05)。在培养7 d后组C的ALP活性大于组A大于组B,差异具有统计学意义(p<0.05)。培养24 h时组C细胞伸展良好,有大量伪足向四周伸展。结论:液体中保存可以减少种植体活性丧失,在NaOH中保存效果最好。

Abstract: Objective: To investigate the effects of different storage methods on adhesion and proliferation rate of osteoblasts on the surface of titanium implant. Methods: The titanium plates were modified with sandblasted and acid-etching technology and divided into three groups according to the storage methods. Group A was in the airtight container, group B was in the steaming water (ddH2O), and group C was in the sodium hydroxide (NaOH) solution for 4 weeks. Adhesion and proliferation of osteoblasts at different time points on the surface of titanium were evaluated by MTT assay, alkaline phosphatase activity assay, and laser scanning confocal microscope. Results: After 1 hour and 3 hours culture, the number of cells on group B and group C surface were higher than that on group A (P<0.05). After 1, 4, 7 days culture, the number of cells on group C was higher than those on other two groups (P<0.05). Under laser scanning confocal microscope, osteoblasts were found to spread better on group C surface. After 7 days culture, alkaline phosphatase activity of cells on group C was higher than those on group B and group A (P<0.05). Conclusion: The titanium plates preserved in the liquid was beneficial to maintain the biological activity. Sodium hydroxide solution might be the optimal way for implant storage.
