红景天苷对成骨细胞的作用及相关分子机制研究

Effect of Rhodiola Rosea Glycosides on Function of Osteoblasts and Related Molecular Mechanism

摘要: 目的:研究红景天苷(SAL)对人成骨样MG-63细胞增殖、分化和HIF-1α依赖的VEGF表达的影响及相关分子机制。方法:采用MTS法、流式细胞术分析、RT-PCR、ELISA、Western blot等技术观察SAL对MG-63细胞增殖、分化和HIF-1α依赖的VEGF表达的影响,进一步探讨其相关分子机制。结果:SAL通过调控MG-63细胞的周期分布及上调Runx2和Osterix表达促进成骨细胞的增殖和分化。另外,SAL可明显上调下游靶基因VEGF的表达。结论:SAL可显著促进成骨样MG-63细胞的增殖、分化以及HIF-1α依赖的VEGF的表达,进而促进骨和血管新生。

Abstract: Objective: To study the effects of salidroside (SAL) on proliferation, differentiation, mineralization, and the expression of VEGF depending on HIF-1α of MG-63 cells and related molecular mechanisms. Methods: To study the effects SAL on proliferation, differentiation, mineralization, the expression of VEGF depending on HIF-1α, and related molecular mechanisms, the cell vitality assay, Flow cytometry analysis, semi-quantitative reverse transcription polymerase chain reaction, ELISA and Western blot were used. Results: SAL promoted the proliferation and differentiation of MG-63 cells through regulating the distribution of cell cycle, the mRNA and protein levels and the expression of HIF-1α. SAL could also increase the VEGF production of downstream target genes of MG-63 cells. Conclusion: In vitro experiment study shows that SAL can promote proliferation, differentiation, mineralization, the production of HIF-1α and downstream target genes VEGF of MG-63 cells to promote the formation of new bones and vessels.
