Genes Related to Fibroblast Proliferation in Raw 264.7 Cells with Overexpressed Mutant SH3BP2.

Abstract: Objective: To find genes related to fibroblast proliferation by comparing gene expression profile in RAW 264.7 cells with overexpressed wild type and mutant SH3BP2 gene. Methods: Total RNA in RAW 264.7 cells with overexpressed wild type or mutant SH3BP2 gene were extracted respectively by Trizol and analyzed by microarray to compare differences in gene expression profiles. Database search and real time PCR were performed to verify the most significant related genes in fibroblast proliferation. Results: Microarray data indicated that 734 genes show significant expression differences between two cell lines, among which, 313 genes were up-regulated and 421 down-regulated in cells with mutant SH3BP2 compared with wild type. Following investigation with database search and real time PCR, 4 genes were confirmed the top relevant genes to fibroblast proliferation: CXCL10, IL10, Tnfrsf13b and Pf4. Conclusion: Our study showed expression profiles differences between two cell lines. Four genes were verified to be closely related to fibroblast proliferation and might be the potential key component in the mechanism of Cherubism. This study provides us a bright new window into further study of the disease.
