Detection of NLRP3 mRNA Expression in Periodontitis Patient's Own Tissue Nucleic Acid

Abstract: Objective: To determine the mRNA expression of NLRP3,NLRC4, and related genes IL-18,IL-1β,Caspase-1 mRNA in periodontitis patient's own tissue nucleic acid. To define a regulatory role of NLRP3 in periodontitis. Methods: Inflammatory periodontal tissue samples of chronic periodontitis patients were taken in periodontal flap surgery,and healthy gingival tissue samples were taken from orthodontic patients during tooth extractions.Total RNA from gingival tissue was extracted and reverse transcribed into cDNA, cryopreserved for further use. Real-time PCR was used to detect the mRNA expression of NLRP3,NLRC4, related genes IL-18,IL-1β, and Caspase-1 mRNA. Results: The mRNA levels of NLRP3,IL-18,IL-1β,Caspase-1 in the periodontitis patient's own tissue nucleic acid were higher than healthy gingival tissue. However, the mRNA expression of NLRC4 was not significant different between two groups. Conclusion: The high expression of NLRP3 and related genes IL-18, IL-1β, Caspase-1 mRNA in periodontitis patient's own tissue nucleic acid suggested that NLRP3 has an influence on the pathogenesis of periodontitis.
