Expression of Transcription Factor XBP1 in Oral Squamous Cell Carcinoma.

Abstract: Objective: To detect the expression and explore the role of X box binding protein 1 (XBP1) in oral squamous cell carcinoma (OSCC). Methods: Real-time PCR was performed to detect the expression of X box binding protein 1 splicing (XBP1-s) in 50 cases of fresh primary OSCC samples and the corresponding matched epithelial tissues. The tissue microarray technique and immunohistochemistry were used to detect XBP1 protein expression in 100 cases of OSCC samples, and each case included the adjacent normal epithelial, primary tumor center area, invasive front and lymph node metastasis. Results: The relative expression of XBP1-s mRNA with more than 2-fold increase in OSCC tissue was closely related with lymph node metastasis (P<0.05). Immunohistochemistry staining showed that the expression levels of XBP1 protein in the invasive front and lymph node metastasis were higher compare with that in the epithelial and tumor center (P<0.05). Additionally, XBP1 protein expression in lymph node metastasis was significantly higher than that in invasive front (P<0.05). Moreover, XBP1 protein expression was significantly correlated with pathological grade of OSCC tissues (P<0.05). Conclusion: XBP1 might play an important role in OSCC invasion and promote tumor metastasis.
