LP-17多肽对牙龈卟啉单胞菌脂多糖致牙周炎进展的作用

Effect of LP-17 on Porphyromonasgingivalis Lipopolysaccharide Caused Periodontitis.

摘要: 目的: 探究髓系细胞触发受体-1(TREM-1)的抑制剂LP-17多肽对牙龈卟啉单胞菌脂多糖(P. gingivalis-LPS)诱导的小鼠牙周炎进展的作用。方法: 在C57BL/6小鼠上颌第二磨牙龈沟内隔日注射P. gingivalis-LPS(10 μg),构建实验性牙周炎模型。每日给予腹腔注射LP-17(10 mg/kg),对照组注射PBS。14 d后处死小鼠,收集牙槽骨,micro-CT测量牙槽骨的吸收量,HE染色检测牙周组织的病理学表现,TRAP染色检测牙周组织中破骨细胞的数目,免疫组化检测牙周组织中TREM-1的表达情况。结果: 龈沟内注射P. gingivalis-LPS成功诱导了小鼠的实验性牙周炎。LP-17多肽能够减轻牙周组织的炎症细胞浸润和胶原纤维变性程度,抑制牙周组织中TREM-1的表达,减少破骨细胞数量和牙槽骨吸收量。结论: LP-17多肽可以减轻P. gingivalis-LPS诱导的牙周炎症反应,延缓牙周炎进展。

Abstract: Objective: To explore the effect of LP-17, an inhibitor of triggering receptors expressed on myeloid cells-1 (TREM-1) on porphyromonasgingivalis lipopolysaccharide (P. gingivalis-LPS) induced periodontitis in mice. Methods: P. gingivalis-LPS was injected into maxillary second molar sulcus of C57BL/6 mice to construct experimental periodontitis model. Mice were treated with LP-17 peptides by intraperitoneal injection every day, and PBS was set as the control. After 14 days, all mice were sacrificed. Micro-CT was used to measure the alveolar bone absorption. HE staining was used to assess pathological alteration and TRAP staining was used to detect osteoclasts in periodontium. In addition, the expression of TREM-1 was determined by immunohistochemistry. Results: P. gingivalis-LPS induced experimental periodontitis in mice. LP-17 reduced the degree of collagen fibrosis and inflammatory cell infiltration. Additionally, LP-17 inhibited the expression of TREM-1 in periodontal tissue. Furthermore, LP-17 treatment notably decreased the number of osteoclasts and alveolar bone absorption. Conclusion: LP-17 peptides alleviated the inflammatory response in periodontal tissue, thus arrested the progress of periodontitis.
