负载D-缬氨酸水凝胶对大鼠实验性牙周炎的影响

Effects of D-valine Loaded Hydrogel on Rat Periodontitis

摘要: 目的: 探讨负载D-缬氨酸水凝胶对大鼠实验性牙周炎及相关炎症因子表达的影响。方法: 将30只SD大鼠分为阴性对照组(A组)和牙周炎模型组(B组),再在模型组基础上给予不同药物,分为派丽奥组(C组,阳性对照组)、负载D-缬氨酸水凝胶组(D组)、空白水凝胶组(E组),共分为5组,每组6只。成功构建牙周炎模型后,测量各组给药后牙龈指数(GI)、菌斑指数(PI)、牙周袋深度(PD),CBCT测量牙槽骨吸收值(ABL),HE染色观察牙周组织病理变化,ELISA检测IL-1β、IL-6、IL-10、TNF-α水平。结果: 模型组牙周指数,IL-1β、IL-6、TNF-α均升高,IL-10降低;治疗后C、D组GI、PI、ABL明显低于模型组(P<0.05),3组PD值与模型组比较差异无统计学意义(P>0.05);D组IL-1β、IL-6、TNF-a水平明显低于模型组(P<0.01),进一步治疗D组IL-6、TNF-α与A组比较差异无统计学意义(P>0.05),D组IL-10明显高于模型组(P<0.01)。结论: 负载D-缬氨酸水凝胶可降低炎症因子水平,减少牙槽骨吸收,对改善牙周炎具有较好疗效,可能与抑制牙周致病菌有关。

Abstract: Objective: To investigate the effect of D-valine hydrogel on experimental periodontitis and related inflammatory factors in rats. Methods: Thirty SD rats were divided into five groups: negative control group (group A), periodontitis model group (group B), the periodontitis with PaiLi (group C, positive control group), the periodontitis with D-valine hydrogel (group D), and the periodontitis with blank hydrogel (group E). The gingival index (GI), plaque index (PI), periodontal pocket depth (PD), and alveolar bone absorption (ABL) were measured by CBCT. Pathological changes of periodontal tissues were observed by HE staining, and the levels of IL-1β, IL-6, IL-10, and TNF-α were detected by ELISA. Results: The periodontal index, IL-1β, IL-6, and TNF-a in the model group were all increased and IL-10 was decreased. The GI, PI, and ABL in the group C and D were significantly lower than those in the group B (P<0.05), and the PD values in the groups C, D, and E were not significantly different from those in the group B (P>0.05). The IL-1 β, IL-6, and TNF-a levels in the group D were significantly lower than those in the group B (P<0.01), while the IL-6, TNF-a levels in the group D were not significantly different from those in the group A (P>0.05). And the level of IL-10 in group D was significantly higher than that of group B (P<0.01). Conclusion: D-valine hydrogel can reduce inflammatory factor level, reduce alveolar bone resorption, and improve periodontitis, which may be related to periodontal pathogens.
