1型糖尿病小鼠下颌密质骨厚度特点的研究

摘要: 目的:运用Micro-CT对1型糖尿病小鼠颌骨密质骨厚度的改变进行研究。方法:选用6周龄小鼠20只,随机分为对照组和糖尿病组,糖尿病组按50 mg/kg剂量连续5 d腹腔内注射链脲佐菌素建立1型糖尿病小鼠模型,对照组腹腔注射枸橼酸盐缓冲液。饲养12周后处死小鼠,取全血进行血清钙磷检测;通过Micro-CT扫描小鼠下颌骨比较糖尿病组和对照组密质骨厚度的特点。结果:糖尿病组血清钙水平较对照组降低,血清磷水平升高,钙磷乘积下降。Micro-CT观察到糖尿病组小鼠密质骨厚度较对照组明显变薄,两组密质骨厚度的比较差异有统计学意义(P<0.01)。结论:本实验通过链脲佐菌素诱导的1型糖尿病小鼠密质骨结构发生了改变,Micro-CT结果显示糖尿病状态下颌骨密质骨厚度明显变薄。

Abstract: Objective: To observe the mandible cortical bone thickness changes in type 1 diabetes mellitus mice using Micro-CT. Methods: Twenty 6-week-old mice were randomly divided into control group and diabetes mellitus group. The diabetes mellitus group received intraperitoneal injection of 50 mg/kg streptozotocin for 5 days to establish a type 1 diabetes mellitus model, and the control group received intraperitoneal injection of citrate buffer. Then the mice were sacrificed after 12 weeks. The serum calcium and serum phosphorus levels were detected. Micro-CT technique was used to observe the cortical bone thickness changes of the mandible in type 1 diabetes mellitus mice and control group. Results: Compared with control group, the serum calcium level in diabetes mellitus group was decreased, the serum phosphorus level was increased, and the calcium-phosphorus product level was decreased significantly. Micro-CT showed that the thickness of mandible cortical bone in the interest region of type 1 diabetes mellitus mice was significantly decreased in contrast to that of control group (P<0.01). Conclusion: The streptozotocin -induced type 1 diabetes mellitus mice model suggested that the three-dimensional structure of the cortical bone were changed significantly. Micro-CT showed the thickness change of the cortical bone was more obvious.
