Jumonji Domain-containing Protein 3 Regulates the Inflammatory Response of Human Periodontal Ligament Cells

摘要: 目的: 研究三甲基化组蛋白H3赖氨酸K27(histone 3 lysine 27 trimethylation, H3K27me3)的去甲基化酶Jumonji结构域蛋白3(Jumonji domain-containing protein 3, Jmjd3)在人牙周膜细胞炎症反应中的调节作用。方法: 分别使用牙龈卟啉单胞菌(Porphyromonas gingivalis,p.g)的脂多糖(lipopolysaccharide,LPS)和肿瘤坏死因子肿瘤坏死因子-α (tumor necrosis factor-α,TNF-α) 刺激人牙周膜细胞,建立炎症模型。通过实时荧光定量聚合酶链反应(quantitative real-time polymerase chain reaction, Q-PCR)和免疫荧光检测Jmjd3的表达情况。后通过沉默Jmjd3,在Q-PCR和Western blot确定沉默效果后对其可能的下游靶基因进行初步筛选。结果: Jmjd3在炎症的人牙周膜细胞中表达会上调。沉默Jmjd3后,炎症介导的白细胞介素-6和白细胞介素-12b的上调受到抑制。结论: Jmjd3参与人牙周膜细胞炎症反应,并能够调节白细胞介素-6、白细胞介素-12b的表达。

Abstract: Objective: To investigate the role of the histone 3 lysine 27 trimethylation (H3K27me3) demethylase Jumonji domain-containing protein 3 (Jmjd3) in the regulation of the inflammatory response of human periodontal ligament cells (HPDLs). Methods: HPDLs were stimulated with lipopolysaccharide (LPS) from p.g or tumor necrosis factor-α. The expression of Jmjd3 in HPDLs was examined by quantitative real-time polymerase chain reaction (Q-PCR) and immunofluorescence. After confirmed the depletion of Jmjd3 by Q-PCR and Western blot, potential target genes were selected by silencing Jmjd3. Results: Q-PCR and immunofluorescence revealed that the expression of Jmjd3 was increased in inflamed HPDLs. Knockdown of Jmjd3 led to the suppression of inflammation-induced up-regulation of interleukin-6 and interleukin-12b. Conclusion: Jmjd3 regulated the activation of interleukin-6 and interleukin-12b in the inflammatory response of HPDLs.
