IL-24 Might Inhibit Migration and Invasion Abilities of Oral Squamous Cell Carcinoma Cells through CXCL12/CXCR4 Signal Axis.

Abstract: Objective: To investigate the effect of lentivirus-mediated IL-24 gene transfection on migration and invasion of oral squamous cell carcinoma (OSCC) cells and the effect on CXCL12/CXCR4 axis signal. Methods: OSCC cell lines were cultured and divided into combined treatment group, IL-24 transfected group, inhibitor group, and blank control group. After transfected culture for 48h, the levels of IL-24 in the cytosol were detected by using ELISA, and cells migration, invasion abilities, and apoptosis in different groups were detected. The expressions of IL-24, CXCL12, and CXCR4 genes and proteins in cells were detected by using real-time quantitative PCR and Western blot. Results: The levels of IL-24 in the supernatants of combined treatment group and IL-24 transfected group were significantly higher than those of inhibitor group and blank control group (P<0.05). The numbers of cell migration and invasion in the combined treatment group were less than those in the IL-24 transfected group, inhibitor group, and blank control group, and those in the IL-24 transfected group and inhibitor group were less than those in the blank control group (P<0.05). The expression levels of IL-24 mRNA and proteins in the combined treatment group and IL-24 transfected group were higher than those in the inhibitor group and blank control group (P<0.05). The expression levels of CXCL12 and CXCR4 genes and proteins in the combined treatment group and IL-24 transfected group were lower than those in the inhibitor group and blank control group (P<0.05). Conclusion: Up-regulation of IL-24 could inhibit cell migration and invasion of OSCC. It
