Effects of Chondroitin Sulfate Proteoglycan on MC3T3-E1 Cell Line Mineralization.

Abstract: Objective: To observe the effect of chondroitin sulfate proteoglycan (CSPG) on the alkaline phosphatase (ALP) activity and calcium deposition quantity in pre-osteoblast cell line MC3T3-E1 mineralization process. Methods: MC3T3-E1 cells were cultured with different concentrations of CSPG(0,5,10,15 μg/ml)in induction medium,without CSPG as control group,the other three groups as the experimental group. Cells were cultured for 14 days by ALP staining and activity assay,21 days by alizarin red staining and calcium deposition assay. Results: Compared to the control group,ALP staining and alizarin red staining showed a decreasing trend with the increasing of CSPG concentration in the experimental group;The groups was statistically significant difference between the amount of calcium deposition (P<0.05), no significant difference in ALP activity. Conclusion: In the experimental period,CSPG inhibited calcium deposition of MC3T3-E1 cells in a dose dependent manner.
