Effects of Autologous Concentrated Growth Factors Extract on MC3T3-E1 Cells

Abstract: Objective: To evaluate the effect of the concentrated growth factors (CGF) extract on the proliferation and differentiation of MC3T3-E1 osteoblasts. Methods: The experimental group was cultured with α-MEM containing CGF, while the control group was α-MEM. The proliferation of the cell was detected by CCK-8, the activity of alkaline phosphatase was detected by ALP, and the expressions of runt-related transcription factor-2 (Runx2) and osterix (Osx) genes were quantified by real-time PCR. Results: CCK-8 showed that the experimental groups with different concentrations of CGF had different proliferation and the 10% CGF group was higher than other groups. When 10% CGF was used as experimental concentration, the ALP activity of experimental group was higher than that of control group after 3, 5, and 7d. RT-PCR results showed that the expressions of Runx2 and Osx in experimental group significantly enhanced. Conclusion: The growth factors released by CGF can promote the proliferation and differentiation of MC3T3-E1 osteoblasts.
