缓释型柠檬苦素纳米聚合物抗生物膜作用的研究

Anti-biofilm Effect of Sustained Release Limonin Polymer Nanopolymer

摘要: 目的:研究载柠檬苦素聚合纳米粒(LIM/PLGA-PEG)的抗生物膜性和酸性条件下的缓释性能。方法:制备载柠檬苦素聚合物纳米粒,并对其进行表征。测定其抗变形链球菌和抗生物膜性能。评价LIM/PLGA-PEG的释放曲线(物质毒性)和细胞毒性。结果:LIM/PLGA-PEG显示出抗变形链球菌特性(MIC为1.56 mg/mL)。亚抑制浓度的LIM/PLGA-PEG可降低92%的生物膜形成。在中性和酸性环境下检测LIM/PLGA-PEG在12 h内的缓慢释放。载药纳米粒的IC50为15.63 μg/mL。结论:含柠檬苦素纳米粒具有抗龋活性和酸性环境下的缓释作用, 有望用于控制龋齿的发生和发展。

Abstract: Objective: To study the anti-biofilm effect and sustained release properties of LIM/PLGA-PEG. Methods: Limonin loaded polymer nanoparticles were prepared and characterized. The anti-Streptococcus mutans and anti-biofilm properties were determined. LLIM/PLGA-PEG release curve (material toxicity) and cytotoxicity were measured. Results: LIM/PLGA-PEG showed anti Streptococcus mutans (MIC=1.56 mg/mL) property. Subinhibitory concentrations of these nanoparticles reduced the biofilm formation by 92%. The slow release of LIM/PLGA-PEG was detected in neutral and acidic environment within 12 hours. The IC50 of the nanoparticles was 15.63 μg/mL. Conclusion: The limonin nanoparticles have anti caries activity and sustained-release effect in acidic environment, which is expected to be used in the control of dental caries
