Relationship between Lymph Node Metastasis and Long Non-coding RNA HOTAIR in Tongue Squamous Cell Carcinoma.

Abstract: Objective: To investigate the correlation between Hox transcript anti-sense intergenic RNA (HOTAIR) and lymph node metastasis in tongue squamous cell carcinoma (TSCC). Methods: The expression of HOTAIR was detected in 19 TSCC tissues and 19 matched nonmalignant tissues by using Real-time quantitative PCR, and its correlation with TNM stage was analyzed. The migration and invasion abilities were evaluated in CAL27 and SCC9 cell lines after silencing the expression of HOTAIR. Results: 26.3% of TSCC tissues showed a higher expression of HOTAIR compared to the matched nonmalignant tissues (P=0.0169), and all these patients underwent lymphatic metastasis. Patients without lymphatic metastasis showed a low expression of HOTAIR (P<0.001). After silencing the expression of HOTAIR, the tumor cells' ability of migration and invasion was decreased (P<0.001). Conclusion: High expression of HOTAIR may be an indicator of lymphatic metastasis in TSCC.
