Preliminary Study on Role of Inducible Regulatory T Cells 35 in Etiology of Oral Lichen Planus

Abstract: Objective: To study the expression of inducible regulatory T cells 35 (iTr35), regulatory T cells (Treg), and interleukin 35 (IL-35) in peripheral blood, and to explore their role in the pathogenesis of oral lichen planus (OLP). Methods: A total of 35 OLP patients diagnosed in the Affiliated Hospital of Guizhou Medical University from April 2016 to November 2017 were selected. During the same period, 25 healthy volunteers were selected as normal control group. The level of IL-35 in peripheral blood was detected by ELISA. Its relationship with iTr35 cells was analyzed. Among the cases collected above, 27 OLP patients and 16 normal controls were selected. Flow cytometry was used to detect the proportion of peripheral blood iTr35 and Treg cells, and the relationship between two cells was analyzed. Results: Compared with the normal control group, the number of Treg and iTr35 cells and the expression level of IL-35 in OLP were significantly increased, and the differences were statistically significant (P<0.05). Spearman correlation analysis showed that iTr35 cells were positively correlated with Treg cells (r=0.385, P=0.048) and CD4+Foxp3+Ebi3+p35+ cells (T cells that produced IL-35 in Treg cells, r=0.723, P<0.001). While the expression level of IL-35 was not correlated with iTr35 cells (r=-0.173, P=0.454). Conclusion: The high level of IL-35 expressed in OLP serum provides the possibility for the generation of iTr35 cells. Treg cells participate in the generation of iTr35 cells. The increased proportion of iTr35 cells may play an important role in the occurrence and development of OLP.
