Comparison of Viability and Apoptosis of Gingival Fibroblasts in the Patients with Chronic Periodontitis and Healthy People.

Abstract: Objective: To investigate the differences in the proliferation and apoptosis of human gingival fibroblasts (HGFs) between patients with chronic periodontitis and healthy people and provide experimental evidence for the susceptibility of chronic periodontitis. Methods: HGFs were obtained from patients with chronic periodontitis and healthy people and cultured in vitro. Cell activity between two groups was detected by MTT assay. Real-time PCR was used to detect the expression levels of anti-apoptotic gene Bcl-2 and pro-apoptotic gene Bax in HGFs from the two groups, and the ratio of Bcl-2 to Bax was compared. Results: Compared with the healthy group, the cell activity of the HGFs in chronic periodontitis group decreased significantly after 4 days culture (P<0.05). The ratio of Bcl-2/Bax significantly decreased after 2 days and 4 days in vitro in a time-dependent manner (P<0.05). Conclusion: HGFs cells from patients with chronic periodontitis show low cell activity and high cell apoptosis, which indicates the susceptibility of periodontitis.
