Effects of Two-dimensional and Three-dimensional Culture on Biological Characteristics of Human Tongue Squamous Cell Carcinoma TCA8113 Cell Lines.

Abstract: Objective: To compare the effect of 2D and 3D culture on the cell proliferation, organization form, and activity of lactate dehydrogenase of TCA8113 tongue squamous cell carcinoma cell line, and to provide methods for the culture of tongue squamous carcinoma cells in vitro. Methods: The morphology of TCA8113 cells was observed by inverted microscope and HE staining. MTT method was used to observe the proliferation of cells in two types of culture. ELISA was used to detect the changes in LDH activity. Results: Compared with the 2D culture system, the cells in the 3D culture system maintained a good proliferation status in the long period, and the activity of LDH was significantly higher than that of 2D culture. Conclusion: Alginate gel 3D culture system was better than 2D culture system for to reflect the biological characteristics of TCA8113 cells.
